When I try to add boxes to my HOME page, Nick's video tells me to choose "Home" as the location for the boxes, however I am only given the "default" choice. HOME is not listed. Have I perhaps not set my HOME page correctly?

That is true, Default Page exists as a template page type. Are you adding the Boxes section to Page Templates or Content Area? Also, did you check to make sure that Boxes are not set to hide in the PageLines Meta Settings (in the WP page editor)?
