How/where/can you change the content that is auto populated into a tweet when someone using the Twitter sharing icon at the bottom of a post? Right now it appears to pull from the post Title ... is that correct?

I think that will require that you install and activate a different Twitter sharing plugin that allows you to customize how tweets are formatted. Try searching for one in: http://wordpress.org/extend/plugins/

It uses the URL of the post.
